Background The protein encoded by this gene belongs to the ser/thr protein kinase family. It is the catalytic subunit of the 5'-prime-AMP-activated protein kinase (AMPK). AMPK is a cellular energy sensor conserved in all eukaryotic cells. The kinase activity of AMPK is activated by the stimuli that increase the cellular AMP/ATP ratio. AMPK regulates the activities of a number of key metabolic enzymes through phosphorylation. It protects cells from stresses that cause ATP depletion by switching off ATP-consuming biosynthetic pathways. Alternatively spliced transcript variants encoding distinct isoforms have been observed. Western blot analysis of extracts from K562 cells using db15794 at 1:1000.
Immunofluorescence analysis of MCF-7 cells labelling AMPK alpha 1 with db15794.

The cells were fixed with cold 100% methanol (10min, 4℃) and blocked in 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween 20 for 1h. The cells were then incubate with db15794 (1:100) at room temprature for 1h, followed by a further incubation at room temperature for 45min with Goat Anti Rabbit lgG (H+L)-AF488 (db10005, shown in green). Nuclear DNA was labeled in blue with DAPI.
