Background
NF-kappa-B is a ubiquitous transcription factor involved in several biological processes. It is held in the cytoplasm in an inactive state by specific inhibitors. Upon degradation of the inhibitor, NF-kappa-B moves to the nucleus and activates transcription of specific genes. NF-kappa-B is composed of NFKB1 or NFKB2 bound to either REL, RELA, or RELB. The most abundant form of NF-kappa-B is NFKB1 complexed with the product of this gene, RELA. Four transcript variants encoding different isoforms have been found for this gene. Western blot analysis of extracts from Jurkat, C6 cells using
db13524 at 1:1000.
-
Immunofluorescence analysis of HeLa cells labelling NF-KB p65 with
db13524.
The cells were fixed with cold 100% methanol (10min, 4℃) and blocked in 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween 20 for 1h. The cells were then incubate with
db13524 (1:100) at room temprature for 1h, followed by a further incubation at room temperature for 45min with Goat Anti Rabbit lgG (H+L)-AF488 (
db10005, shown in green). Nuclear DNA was labeled in blue with DAPI.
Control: Secondary antibody only.
